Some drugs for the treatment of cancer have been identified due to serendipity (e. g. thalidomide, rapamycin) However, in practise, there are two main approaches used in modern cancer drug discovery rational drug design and high throughput screening. Rational drug design involves designing and synthesising compounds based on the known or predicted structure (e. g. crystal structure) of a single specific target or interaction. The agent (and its structure if relevant) and/or the target may be unknown. Provides large-scale, efficient measurement of the effects of agents or conditions in biological or chemical assays. Important tool used in drug and biomarker discovery. Robotics and automation platforms -> can"t really dispense into the tiny wells in big plates. Plate-reader-based assays (chemical, biochemical, enzymatic) -> we did mtt assay, plate-reader based assay. Need to have a plan -> do the same thing each time. Streamlined data analysis and bioinformatic pipelines for large datasets.
