To stain something it must be demineralised because you can"t cut thin sections with a microtome because it is too hard. Demineralise mature enamel, it is lost and appears as a space. If you see enamel as a space in histological stained sections mature because in process of demineralisation all enamel has been lost. If enamel is still there and accepts staining immature. Initially deposited enamel is high in protein and water and low in minerals.
