Lecture 3: Behaviour of chromosomes + alleles
The basic principles of the PCR (polymerase chain reaction)
Add: DNA sequence to be amplified, deoxyribose sugar, enzyme, primer
Need to know something about sequence to design primer
1. Use heat to separate DNA strands
2. Cooling: primers added to the ends of region of interest
3. Taq polymerase adds new bases complementary to template
Genes and Alleles
Genes can be found in alternative forms --> allele
Wild type = most frequent
Alleles arise due to mutations (change DNA sequence)
Chromosome:
After DNA replication - chromatids = identical
Homologous chromosomes: same genes but different alleles
Diploid:
oTwo of each type of chromosome
oTwo similar chromosomes = homologous
Haploid:
oOne copy of each chromosome
Mutations:
Aneuploidy: +/- chromosomes but not a set
oNon disjunction at meiosis at anaphase 1 or anaphase 2
Euploidy: extra sets
MEIOSIS:
Segregation of alleles
Independent assortment of chromosomes
Crossing over and recombination of alleles
Segregation of alleles
Anaphase 1 and Anaphase 2
