Add dna sequence to be amplified 4 deoxyribonucleotide triphosphates, deoxyribose sugar, enzyme and primer. Need to know something about sequence to design the primers. Primers can be designed to only find one complementary piece in the entire genome (amplifies the specific sequence) Primers complementary to the ends of the region of interest are annealed when cooling occurs (60-65celsius) Each base has 1/4 chance of being there -> a chance is 1/4, c 1/4, t. Chance of finding that region complementary in dna is (1/4)^20. Adds new bases complementary to the template dna. Sequence can be amplified millions of times after several cycles of pcr. Genes can be found in alternative forms - alleles. Most frequently occurring allele in a population may be referred to as a wild type (not used for human alleles) Same gene loci, but not necessarily same allele. Fungi spend most of life cycle as haploid.
