Week 7 trafficking, life and death of the cell. Use an example to demonstrate how impairing mitosis can be used in therapy. Used for treatment of breast, ovarian, lung, bladder, prostate, melanoma, oesophageal and other solid tumour cancers. Compound act against cdk-cyclin are being evaluated for various cancers. Isolated from pacific yew tree (rare) : but now made in bioreactors using plant cells. Paclitaxel treated cells (low concentrations) are often arrested at metaphase due to defects in spindle mt dynamics. Spindle assembly checkpoint is activated since some kinetochores are not attached. Thought to cause mitotic arrest but the concentration in tumours varies and multipolar divisions occur instead. Describe how defects in mitosis can lead to disease. The presence of an abnormal number of chromosomes in a cell. Describes the karyotype of a single somatic cell when it has anything other than two copies of each chromosome. Does not distinguish between whole-chromosome gain and loss.
