Gsprs are integral membranes - 7 transmembrane domians g-proteins cannot leave the membrane, but can slide along. Heterotrimeric - 3 proteins that are not related to each other, but come together to form one unit. The alpha and gamma proteins are anchored to the membrane, beta sits neatly between them. Gdp is the ligand - chnages the affinity to high affinity. When the g-protein is connected to gtp, it results in 3 portein units being activated. In humans, 400 different gpcr genes for smell, in mice, there are 800 oi these. The largest family of transmembrane receptors - approx. 1000 in the human genome, more in other animals. Act as receptors for proteins, hormones, neurotransmitters, odours, light, many are. Effectors are diverse, and act through diverse second messengers to elicit diverse cellular responses. Some that will indicate light has be experienced. Some will cause other things to be released. 30-50% of current pharmaceutical agents work through gpcrs.
