Oral corticosteroid prednisolone or prednisone mtor inhibitors: sirolimus, everolimus. Block il-2 mediated transduction pathways preventing movement of activated t-cell from g1 to s phase of proliferation large pk variability between pts so need close tdm (therapeutic drug monitoring) Ae"s: hypertension, hyperlipidaemia, thrombocytopenia, impaired wound healing. Antagonists at il-2 receptor on t-cells, block binding of il-2 preventing t- cells from becoming activated by il-2 and proliferating. Adverse effects: few reported, no known drug interactions. Reduce incidence and severity of early acute rejection without increasing incidence of infections or adverse effects. High dose steroids methylprednisolone 1g (iv) for 3 days.
