Toxicity depends not only on the inherent properties of a chemical, but also on size of administered dose o dose = total amount or mass of material to which an individual is exposed (e. g miligrams or grams) o dosage = defines amount of material in relation to a characteristic of recipient (e. g body weight) (e. g miligrams/kg) Often can involve: genetic traits for abnormal response, genetic traits for abnormal metabolism o hypersensitivity responses: Major routes for toxicant entry: o gastrointestinal tract (ingestion) o lungs (inhalation) o skin (topical) o other e. g parenteral. Most systemic toxicants damage a few target organs" o may be due to pharmacokinetic factors (e. g preferential delivery to target tissue) o or target tissues express enzymes that form electrophilic" metabolites (electron deficient) Cns effects probably more common than effects on other organs (i. e. during chemical intoxications) o can produce diffuse encephalopathy with global o dysfunction.
