Genomic: study of genomes including: determination of the dna sequences of genomes, gene expression and protein expression. How to make money from an organism. Identification of genes of potential use in biotechnology (ex/ heat stable enzymes) Requires: template dna, primer (radioactive dna primer), polymerase, deoxynucleotides, one dideoxy terminator nucleotide per lane. Lanes divided by g, a, t, c. If a terminator nucleotide is incorporated into a growing dna strand, synthesis stops. No addition can happen on the 3" end. Random incorporation of terminators will generate short fragments. Refers to a variety of methods for generating large amounts of sequence data quickly. Highly parallel: thousands to billions of sequences collected simultaneously. 2008 = transition from sanger sequencing to nextgen. * watch video by illumina (1) millions of clusters generated on a glass slide. Can tell sequence by sequence of colours. Can get sequencing from both directions (forward and reverse strand)
