Remember that there are regions in the chromatin that are more condensed than others. They are very condensed and don"t do nothing. Take a cloned piece of dna and label it with a fluorochrome and hybridize it, Melt the dna of a cell and add the fluorescent dna probe to the chromosome of this cell. When you hybridize the probes with the nuclei dna, you see spots of hybridization. As different probes bind to the dna, you can see that they form a loop. Knowing the order in the genome where they will bind, it is possible to conclude that these loops form in the chromosome!
