Guidance and migration in the nervous system: introduction. Proper wiring = the right neurons making the right connections. How do we get the neurons at the right place: most neurons are born in the ventricular zone, most migrate radially outwards in an in-to-out pattern. Neurons migrate radially to pattern the cerebral cortex. Neurons migrate by extending a leading process towards the cortex, and the cell body must move after it (2 alternating processes. Spoiler alert: chemical gradients that mediate the polymerisation/depolymerization of the cytoskeleton via machinery we already saw in class. Migrating neurons leading processes are growth-cone like (but not growth cones) If you stain growth cone, you"ll see f-actin and microtubules. Axon outgrowth is driven by microtubule nucleation. Dcx is found in neuronal processes: look at microtubules in the growth cone shows that new microtubules are created there. Probing for eb1 +tip protein shows that there is a local blooming at microtubules.
