Class of enzyme i: citrate synthase (synthase / transferase ) Purpose : to bring in 2c product of glycolysis. 2-step reaction : first a condensation reaction to join the 2 reactants, then a hydrolysis reaction to remove coenzyme a. The prosthetic group required to stabilize the carbanion intermediate formed in step 2 is a positively charged metal ion mn 2+ (since there is already internal resonance, tpp is not helpful). Note: similar to pyruvate dehydrogenase reaction (decarboxylation followed by oxidation) Ketoglutarate dehydrogenase complex (kdh) similar to pdh complex. Same 5 cofactors used (nad + , fad, coa, tpp, lipoate) Active sites are different to accommodate different-sized substrates. Note: gtp can also be produced by this step. Class of enzyme i: synthetase / ligase (named for the reverse reaction) This is the 3rd slp (first two occurs in glycolysis). Recall: slp = redox reaction followed by a group transfer.
