Tbk1 is a key regulator of type i ifn production. Loss of tbk1 has an impact on type 1 ifn induction after viral infection. Tbk1 deficiency is embryo lethal due to high occurrence of hepatic apoptosis. We report here the targeting of ubiquintinated tbk1 to golgi apparatus after rlr or tlr3 stimulation, through interaction with optineurin (optn), a uniquitinbinding adaptor protein. Optn sense k63-linked polyubiquitin chains on tbk1, triggering the formation of tbk1-optn complexes, in which tbk1 is activated via trans-autophosphorylation. Bluetongue virus neutralizes optn activity at golgi apparatus, decreasing tbk1 activation. Reveals a new function of the golgi apparatus in innate immunity.
