Learning Tasks, Sleep, & Memory
The learning task: The rat is place in one of the compartments, the light and an auditory tone is
presented for 20 seconds in the same compartment the rat is located. Then a shock is
delivered, and all signals terminate at the same time. The rat must learn to cross over to the
other compartment before the shock occurs.
- Rats have very routine lives, so this task is a big learning curve for them.
- If deprivation of REM occurs after the initial learning task then their learning is very
impaired.
100 Trials in 1 Day Procedure (F1)
When rats are given 100 training trials in one day, the REM sleep increases and persists for
days. Is sleep deprivation is applied 1-4 hours after learning subsequent performance on the
task is impaired. REM deprivation outside this 1-4 hour window does not affect subsequent
performance. The period where post-training REM increases are observed, and REM sleep
deprivation impairs performance is called the REM window (also called PSW aka paradoxical
sleep window).
Rats had an increase in REM sleep for 6 days and we don’t know when this tappers off eause
the study ended. Intensity of learning matters and creates different patterns of REM sleep.
50 Trials/Day Over 2 Days
The timing of the REM window is dependent on the training regime.
When 50 trials per day over 2 days (100 trials total) is used, REM increases are observed 1 day
after training (TD1) but not after the second day (TD2). After which large REM increases are
observed. When REM deprivation occurs 9-12 hours and 53-56 hours after training, subsequent
performance is impaired. REM deprivation outside of REM window does not impair
performance.
- There are critical moments and specific times that we need for memory consolidation.
- These results are due to a seemingly important event, sleep engagement when the task
is salient (something emotional, something traumatic).
REM learning and the EEG spectra.
- The sigma band increases in learning condition rats.
Sleep Spindles and Memory: Sleep spindles increased after training 1 day in rats that learned
the task successfully vs. those who did not learn the task.
Hippocampal Cells: code location, initiate memory.
McNaughten and Wilson: looked at activation of place cells and how they interact with other
place cells. Memories are being replayed physiologically when we look at the patterns of
different places. Place cells are reactivated when they are required to remember things or in
the process of remembering things.
