In the first lab we ligated dna with bamhi hindiii: we then took the ligated dna and transformed em into cells, then we checked to see if cells have resistance for both the antibiotics. We can"t really know if colonies in the lb/amp plate has kan resistance and vice versa: we can only conclude when the organism is actually challenged with the stuff. This is called replica plating: we plate 12 colonies from the lb/amp and lb/kan plate (24 in total and on a grid) They are all coming from the ligated colonies (l: you streak colonies from the same l plates (eg/ 12 lb/amp colonies) and smear them onto replica plate (eg/ lb/amp and lb/kan plates, incubate at 37 c overnight. Replica plating tells us the success of lab 1 and parallels results from lab 2.
