Normally, on older instruments, you would get one or a couple peaks. Monoisotopic mass: carbon-12 vs. carbon-13, which makes it one off from the biggest peak. In nature, the natural isotope of carbon (most prominent one) is carbon-12. When a protein is being made in a cell, there is a 1% chance that you will get carbon- You have a population of proteins and when they are digested into peptides, there will be some amount of carbon 13 present somewhere along the peptide backbone, side chains, peptide bonds, etc. The average of that would be if all the isotopic peptides were not separated and you would just get an average mass.
