BIOL 4570 Lecture Notes - Lecture 9: Antigen-Presenting Cell, Antigen Presentation, Mhc Class I
Document Summary
Immunology: class i vs class ii compare & contrast. Evasins block antigen presentation by preventing peptide movement through tap peptide transporter. E19 competes with tapasin and inhibits peptide loading onto mhc i proteins mk3 protein ubiquitinates mhc i degraded by proteasome. Calnexin stabilizes mhc i molecule when it is unbound to peptide. Calreticulin does same thing with help of erp57. Mhc ii pathway: antigen is taken up from ec space into intracellular vesicles. In early endosomes of neutral ph, endosomal proteases are inactive: acidification of vesicles activates proteases to degrade antigen into peptide fragments, vesicles containing peptides fuse with vesicles containing mhc class ii molecules, chaperones. Clip, bound to class ii molecule: clip fits almost all class mhc ii does not have great affinity because it is acting as place holder, sorted into specialized endosomal vesicles only in apcs. Golgi has class ii molecules associated with invariant chain, large diameter circles.
