Two measures to determine how addictive a drug is: kinetics: involves the route of use -> speed of onset (how quickly takes effect) & speed of offset (how quickly it wears off) -> both fast -> highly addictive. 2)dynamics: how efficient is the drug at binding with the receptors or enzymes -> greater efficacy, the greater the addictive potential. In overdose, cocaine constricts blood vessels in the heart => heart attack, seizure, pulmonary oedema (fluid in the lungs) or can rupture the aorta. Heavy use can cause myocardial fibrosis (normal heart muscle is replaced with fibrous tissue, so heart cant pump blood properly) & thromboxane production causing blood clots.
