CHE 113 Lecture Notes - Lecture 10: Luminescence, Phenolphthalein, Luminol
Document Summary
Hemoglobin has peroxidase- like activity (enzymes that accelerate oxidation of organic compounds) Kastle-meyer test- uses phenolphthalein (is it blood?) Take the blood and give it peroxide, which reacts with phenolphthalein and turns pink which indicates it is blood. Reaction of luminol with blood produces a complex which can be seen by luminescence. Very sensitive, up to a 3,000,000 dilution of blood can be seen. Does not interfere with later dna testing. When animals are injected with human blood they form antibodies to the human blood. Can isolate human antiserum (antibodies to human blood) Human antiserum will react only with human blood. Antiserum can (has) been made similarly for many others animals. Works on old (dried for many years) and small samples. Animals (typically mice) are injected with human blood, to stimulate antibody production. At the same time tumor cells are grown in a dish. Antibody cells are fused with tumor cells (called hybidromas)
