Arc antisense blocks consolidation of long-lasting ltp. Arc antisense (as) reversed well established ltp when applied about 160 minutes after the inducing stimulus but not when applied 280 minutes later. This indicates that the synaptic changes are still ongoing at 160 minutes and have been fully consolidated by 280 minutes. Arc sustains actin regulation to allow ltp consolidation. When arc synthesis is prevented there is a corresponding large loss of new actin filaments. This supports the hypothesis that arc contributes to the consolidation of. Without arc present, the earlier increases of ampa receptors in the psd and the changes in spine size are thought to be lost. Note: changes on postsynaptic side of synapse. 1) increase in spine head volume, mediated by increased f-actin (structural protein) 2) increases in synaptic adhesion molecules (cadherin) 3) increase in ampa receptors in postsynaptic membrane. Points 1 & 2 above provide structural stability of the synapse over time.
