Since two strands of dna are complementary, each acts as template for building new strand in replication. Parent molecule unwinds and two new daughter strands are built on base-pairing rules. Replication beings at special sties called origins of replication: where two dna strands are separated, opening up a replication bubble. A eukaryotic chromosome may have hundreds or even thousands of origins of replication. Replication proceeds in both directions from each origin until entire molecule is copied. At the end of each replication bubble is a replication fork, a y-shaped reion where new dna strands are elongating. Hydrogen bonds are broken and replication bubbles are formed. Less energy needed to part the two bases. Moves along double helix, breaking bonds as it goes along. Chemotherapy drugs inhibit human topoisomerase; may lead to cell apoptosis. Dna polymerases catalyze elongation of new dna at replication fork. Most dna polymerases require a primer and dna template strand.
