Aggregatio - no mechanism for clearing up trash. Mutations in proteins may make for earlier age of onset. If autophagy is failing - cells may propagagte disease becayse the expellation of unveonvetional/prion proteins will infect other cells. As we age, we have a decine in atg proteins - this decline causes lysosomes to fail. -> prion like propagation of bad proteins --> cellular stress causes cells to kill itself. Core machinery can be upregulated by startvation which in turn could posible upregulate nutrient rich slelective autophagy. Many types of autphagy - increase in lipids -->cholesterol with aging. Loss of autophagy causes many age related diseases. Evidence by knocking out atg genes in mice and they all got neurodegenerative disorders. Core atg proteins lack may contribute to neurodegnerative disorder. Loss of autophagy in heart can cause many diseases autophagies keep the heart clean ( house keeping role under basal conditions).
