Experiment: lethally irradiated mice took bone marrow from donor mouse and injected into these mice: wait 9 days look in spleen (lymphocyte trafficking) found small. Lecture 2 page 1 colonies growing: bumps represented stem cells injected, cfus-colony forming units of the spleen. Donor mouse provided hematopoietic bone marrow stem cells. Removed spleen and found small bumps (cfus) Didn"t know what stem cells looked like, stuck in whole bone marrow. Could calculate how many stem cells in bone marrow based on how many cfus present. Erythropoiesis- makes erythrocytes thrombopoesis- makes thrombocytes (platelets: white- white blood cell- leukocytes, erythrocyte- red blood cell. Environmental cues: soluble and membrane associated cues cell encounters to tell to differentiate, stromalcells, cytokines combine to give cues. Genetic damage to stem cells recognized and undergo apoptosis creating environment for new cells to be created (transplant cells) receive cues to produce bumps. Bone marrow come stem cells that enter blood.
