BISC403 Lecture 19: Lecture 19
Document Summary
Lecture xix chapter 10: analyzing genomic variation. Today; what came out of it: more than 1800 genes mapped, can find a gene suspected of causing an inherited disease in a matter of days, genetic tests. Hapmap: catalog of common genetic variation, or haplotypes, if we could find the haplotype options. Most polymorphisms don"t influence phenotype: many mutations in codons are silent; don"t change the amino acid, many deleterious mutations disappear from the population through natural selection, codons make up less than 2% of the human genome. Pcr: taq polymerase heat resistant polymerase enzymes, heat enough to unwind. The three steps in each pcr amplification cycle: denature strands, base pairing of primers, and polymerization from primers along templates. Exponential increase in the amount of target dna during pcr. Determining genotype by sequencing pcr products: sickle cell anemia is caused by a snp in the hb gene, genotyping can identify carriers and homozygous individuals.
