The promoter region (-35 and -10) are never transcribed to the rna. Genes are areas where the bases have some meaning (meaning they can code for proteins) So we"re going from one nucleic acid (dna) to a different nucleic acid (rna) and then translate it to amino acids (proteins) To go from dna to rna, there is an enzyme called rna polymerase. Uses dna as a template to create a mrna strand. In prokaryotes, you make the mrna and use it right away. In eukaryotes, the mrna can be cut and sliced together before you use it. The spliceosome is what we call and snrnp (small nuclear ribonucleoprotein complex) This means that splicing is done by a complex involving both rna and protein molecules.
