If you mess with polycomb, fly imaginal disks lose ability to know their role after metamorphosis. One imaginal disk gives rise to wing the other one gives rise to haltere. At imaginal disk stage, wing imaginal stage -> off gene. Prc2 is bound to wing - this is where gene is not expressed. Binding profile in prc = wing, beige = haltere. Bottom line prc2 sits at enhancer region next to gene regardless of whether gene is active or not, so recruitment is insufficient. However, something is different - in actual gene, ther"es a big diff whether prc2 puts. There"s another step, you have to recruit prc2 but that"s not enough, there"s another step where prc2 decides to be marked or not marked. Well we think that rna has something to do with it. Rnaip = immunoprecipitation with edta in solution so rna doesn"t get degraded, but very dirty assay.
