This means that they have defined free energy state in the opposite direction that we would, folded state expect to be -ve but here they have flipped it around. This is a stable protein but +9 can be confusing. Folded state = -ve, but slide has flipped it around. Flip all the signs, remember g should be -ve to be stabilising. Stabilising contributions should be -ve (but they have it as +ve) Mostly experimental and made a bunch of mutations to find relative contributions of each bit. Number of residues * number of kcal per mole per residue. In the end, thisi s what they came up with -15 kcal/mol determines unstable proteins. We add up small contributions, and the are probably happy that they ended up in similar ballpark relatively close to experimental measurement.
