Document Summary
1/22: principles of pharmacology - michela (micky) marinelli. Da neurons - critical for addiction (ne are similar) Mao on mitochondria - degrades some da inside the presynaptic terminal. Drugs mimic/block actions of endogenous compounds (don"t do new things) work to increase/decrease the effect. Semi-synthetic - adjusted compounds (addition/deletion of components) Absorption distribution metabolized excreted: Passive - most prominently (want the drug to be hydrophobic mostly & small, non-polar (degree of ionization), not bound to proteins) If polar - likes water, would be repelled by the membrane bilayer. Nature of drug (acid/base) vs ph surrounding drug. Ion trapping: pka = ph where drug 50% is ionized. Drug is more likely to not be charged. Ha h + + a - weak acid in acidic environ. Acids ionized in more basic environments and can"t go back through (originally goes through due to establishing equilibrium as an acid) Zero-order kinetics - constant amount of drugs eliminated over time.
