Microbial Growth and Nutrition Continued
Streak Plate Technique
Each cell can reproduce to form a separate colony, whether it be visible
growth or a cluster of microorganisms
Used to isolate pure culture
Separate two bacterias from each other by using this technique (if done
correctly)
Steps
Separate petri dish into four quadrants (in your head or use
permanent marker on the back)
In first quadrant, after sterilizing the loop mix the culture with the
loop
Then grab TWO lines from the first quadrant and bring them into
the second quadrant
Continue second quadrant by zig-zagging down the remainder of
this quadrant only
Then grab TWO lines from the second and bring them into the third
quadrant
Continue third quadrant by zig-zagging down the remainder of this
quadrant only
Then grab TWO lines from the third and bring them into the fourth
quadrant
Continue fourth quadrant by zig-zagging down the remainder of this
quadrant only
MAKE SURE TO STERILIZE LOOP AFTER EACH
QUADRANT
Spread Plate Technique
Involves spreading a mixture of cells on an agar surface so that individual
cells are well separated from each other
Usually while in the liquid form (broth)
Steps
Dispense the cells from the liquid onto the medium in the petri dish
Sterilize loop with the flame from the bunsen burner
Use loop to spread cells across the surface of the petri dish
