The cultural methods are selected depending upon the type of microbes. These methods vary with different types of microbes. Two basic methods are used for viable counts of bacteria: standard plate count, most probable number (mpn) Bacterial cells from soil are extracted by appropriate methods and are studied with following methods: streak plate method, spread plate method, pour plate method, serial dilution methods. Used for the isolation of bacteria in pure culture from clinical specimens. One loopful of the specimen is transferred onto the surface of a well dried plate. Spread over a small area at the periphery. The inoculum is then distributed thinly over the plate by streaking it with a loop in a series of parallel lines in different segments of the plate. On incubation, separated colonies are obtained over the last series of streaks. Pour plate is a second method (other than spread plates) to obtain isolated colonies by mechanically diluting a culture (performing a serial dilution)
