Examples: injury, postoperative dental pain, menstrual cramps, headaches, migraines, arthritis, tendonitis, bursitis (fluid filled sacs reduce friction between tendon and bone) Nsaids prevent pain by acting on pns and cns. Daily doses of aspirin or ibuprofen prevent breast, colorectal, lung, prostate, esophageal, stomach and ovarian cancers. Over-expression of cox2 (cyclooxygenase 2) correlates with carcinogenesis and metastases. Potential mechanisms: inhibit pg synthesis, reduce carcinogenesis and metastases. However, nsaids (except aspirin) slightly increases risk of renal cell cancer. Aspirin acetylates and irreversibly inhibits cox-1 in platelets. This inhibits blood clotting for 4 7 days. Because platelets have no dna, further cox-1 cannot be produced. Prevent ischemic strokes due to clots (only aspirin) Prevent heart attacks (only aspirin) only 26% of candidates are on aspirin. By contrast, other nsaids are reversible and effective for only a few hours.
