Paper 4 (purifired protein): microtubule seed attached to coverslip, only observing part attached to coverslip (some seed might fall off -> free floating) Isolates protein complexes -> identify direct protein-protein interaction. 3 moving parts: protein of interest, antibodies, something interacting with protein of interest. Studies interaction between ulk1 & sec 16a. Want to pull down ulk1 using anti-ulk1. Pull down sec16a (interacting with ulk1) at the same time. Bp = blocking peptide: control; specifically block interaction between antibodies & protein of interest. See ulk1 in lysate, but no ulk1 & sec16a in ulk1 ip. Eliminates a specific gene in a certain tissue.
