Basic slide staining techniques: gram stain. Flood the slide with crystal violet solution and let it sit for 1 minute. Rinse off all excess stain with running water. Reflood the slide with gram iodine solution. After rinsing, flood the slide with a discoloring solution for 10-15 seconds. Flood the slide with safranin counter stain for 1 minute. Wash quickly with running water and blot dry with specially designed blotting paper: diff quick stain. Slides are submerged into 3 containers in sequential order. Dip the slide in liquid fixative 10 times. Dip the slide in the stain solution 10 times. Dip the slide immediately in the counterstain 10 times. Rinse for 10-20 seconds under running water and blot dry or airdry. Collection & identification of mites: mites can only be seen via microscope, mange mites. Live and burrow in the skin of dogs and cats. Dull scalpel blade to abrade the skin in an area suspect of mites.
