A hypothesis is a proposal explanation for an observed natural phenomenon. Null hypothesis: there is no significant difference between specified populations, any observed difference being due to sampling or experimental error. If using 4x objective, and the ocular magnification is 10x, the total magnification = 400x. We calibrate the ocular micrometer so that we can measure the cells using the microscope. Field of view decrease in size a the magnification of the image increases. Paper chromatography separates molecules based on their solubility in organic solvents versus water. We characterized the absorption spectra of our isolated plant pigments using a spectrophotometer. Crude extract was applied to a piece of filter paper, filter paper placed in a mixture of organic solvents so that the solvents move up the paper via capillary action. As solvent moves up, it pulls up molecules in the crude extract.
