Today we will be determining bacterial numbers through the utilization of serial dilution and spectrophotometry techniques on escherichia coli cells, which are in the stationary phase. In addition, we will learn about direct and viable counting methods as well as practice the turbidity (spectroscopic) bacterial counting method. These combined techniques are being used because, together, they should result in reliable bacterial count. At the conclusion of this experiment, we expect to find the final dilution of bacteria to be fifty times less concentrated than the original sample. Results of this experiment were favorable, as can be seen in the picture. Our samples became significantly less concentrated in each plate, leading to the isolation of about 26 single colonies at the final dilution. In addition, a graph has been attached to display our record of bacterial density within our samples, shortly after initial dilution.
