Flame the loop (base to tip), allow to cool before touching or placing on culture because will burn you/cause aerosols. Move the tube, not the loop to collect sample. Tap the side when pulling the loop from the nutrient broth. Same except move the loop in zigzag on the agar (not cutting the agar) Mixed culture (many species) versus pure culture (1 species usually grown in slant or broth) Streak plate is a method of isolation and purification. Isolated cells grow in colonies, in clusters, colony forming units (cfus)
