Document Summary

Venom drives mast cell activation which is protective. Mast cell release proteases which digest the venom. Huge drop in body temp if mast cells ko. Omalizumab (humanized anti-ige antibody): binds free ige, leading to clearance, so mast cells don"t get sensitized. Ligelizumab (new version): higher (10-fold) affinity for ige. Enhance mast cell activation in immunity as a vaccine adjuvant. Compound 48/80: activates mast cell and leads to better response to vaccine. If you give vaccine alone mice die quickly. If you give vaccine with the adjuvant that stimulates mast cells, mice survive better. Mast cells release granules with tnf which travel to ln which acts as adjuvants to boost immunity. Describe the major locations of mast cells in the body, how they can be activated and mediators that they produce. Describe in a more detailed fashion ige/antigen-mediated activation of mast cells and its role in allergic symptoms.
