Immunological tolerance
Immunological tolerance is a system for determining which lymphocyte clones will be allowed
to survive.
There are 2 types of tolerance:
- Central (occurs in the primary lymphoid tissues)
Bone marrow (for B cells)
Thymus (for T cells)
- Peripheral
Mediated primarily by regulatory cells
A breakdown in tolerance can lead to autoimmune disease
Genetic factors
- Many autoimmune diseases are linked to the genes responsible for:
1. T cell activation

Activated cytotoxic t cells find their way to the pancreas. B cell-specific ctls kill the b cells. The a a(cid:374)d cells are unaffected: highlights exquisite antigen specificity.
