Studying an axon is not studying synaptic transmission, studying an axon is studying what precedes (comes before) synaptic transmission. Neuromuscular junction uses motor neurons that are coming in onto a motor/muscle fiber causing it to depolarize. On the presynaptic at the terminal there is calcium imaging. You also see a massive influx of calcium presynaptic side. Massive influx of calcium is critical for several reasons. It is carrying the remnants of the action potential. Remnants meaning it"s not sodium carrying that there. The only reason for that is because there"s no sodium channels here. There are inward calcium channels that are carrying positivity inward of calcium which are big molecules. exactly the same way that sodium channels are. Calcium channels are voltage gated in opening, but when they come in they now mobilize calcium binding proteins. Calcium binding proteins (like clathrin) push vesicles towards the inner surface of the presynaptic membrane.
