General chromatography setup: chromatography separates molecules based on their relative affinities for the mobile of stationary phase. What aspect of the proteins is unique to everything else and use it in identifying/doing stuff with it. The column contains the stationary phase beads that are special interact with the protein of interst an adsorbent molecules can stick to the surface of it. In the mobile phase is the buffer water and some kind of salt to keep the protein stable and happy. Molecules are added to the phase and mobile phase is added to the column. Some molecules are slower or faster = fractionation. What initially was a mix of molecules are separated.
