Lecture 5: dna sequencing and the polymerase chain reacion. Need a short fragment of double stranded dna to serve as the template (needs to be denatured) or needs a single strand of dna. Need a primer (usually dna because more stable) nothing would happen without it dntps and dna polymerase. Chemical cleavage: chemicals used for this are very nasty and can cause mutaions in genes. Has the ability to take a short piece of dna and then amplify it many many imes. Because the dna needs to be denatured at high temperatures, dna polymerase would denature each ime. Found a bacteria with polymerase that lived in hot springs called the polymerase taq polymerase.
