The other site right next to it called the p-site, which is the site where the trna that came before this trna is located. The peptide is attached to the trna in the p-site. The trna in the p-site will move to the e site, and it will quickly fall out of the ribosome The sites are made at the junction of the small ribosomal and the large ribosomal subunit. They are half on one subunit and half on the other. Initiation determines the reading frames of the mrna. If it starts into the wrong reading frame, you would get garbage instead of the protein you want. There is only one frame for the code. During initiation the trna goes into the p-site.
