Compound with same structure = key that fit so prevent histamine from attaching to receptor. Ex: chlor-tripolon = antihistamine for allergies - hay fever. *1st drug designed based on understanding of molecular structure. *used strand part of histamine that act as key/fit in receptor & changed rest so wrong key. Problem = caused side effect of drowsiness /sleepiness. So used anti-histamine sleeping property ex: nytol diphenhydramine hydrochloride tablets. *but seldane interact w/ other drugs = ketoconazole [anti-fungi] & caused cardiac attack. Even if effective both hismanal & seldane = withdrawn from fda. *works well but not same extent for people - depend & trial and error. No drowsiness / no interaction with other drugs. Other effective anti-histamine = reactine/zyrtec = cetirizine or claritin = loratidine. If know allergic = avoid substance: anti-histamines. To relieve symptoms of allergic reaction: de-congestants. For congestion in nose symptom only: immunotherapy.
