Document Summary

Inactivated for some reason, leading to over-proliferation of colon cells: people born with this disease only have one wild-type copy of this gene, thus get very frequent colon cancer, apc mutated in all cells (both copies) If had similar disease in the skin, would have lots of little bumps: can take pieces out from the colon and study them; large structure, can look at progression from benign to more and more malignant tumor. If lose one gene, get phenotype of benign tumors. If lose more than that, like p53, may cause mutagenesis: then activation of k-ras leads it to become more and more aggressive as a tumor cell. Involved in wnt pathway: tgf- signaling: mutations in tumor suppressors smad4 or tgf- receptor ii, mutations in genes involved in dna mismatch repair: repair genes that remove little point mutations in the dna.
