Urea = discovered in urine in 1773 by jean rouelle. Urea = believed only made by living organism. *friedrich w hler = considered as father of organic chemistry. Friedrich w hler = first synthesized urea in vitro w/ heat in 1828. Urea cycle = #1 first metabolic pathway defined & discovered by sir hans adolf krebs in 1932. * hans krebs also described cac cycle & won nobel prize for chemistry1 953 for this work. If nitrogen not used = detoxified through urea cycle. 1 cycle = 1 urea made of 2 nitrogen so get rid of 2 nh4. 1st step = mitochondrial matrix of liver cell. Other 4 steps = cytosol of liver cell. Source of ammonium nh4: release from glutamine - non toxic. *not transamination: de-amination of glutamate by gdh. * cps1 = rate-limiting step since use 2 atp. 1st substrate of urea cycle = carbamoyl phosphate.
