Presynaptic boutons are swellings found either along axons or at their tips. They are filled with synaptic (sv) that contain neurotransmitters. Svs fuse with the presynaptic membrane at specialized called presynaptic active zones. The synaptic cleft contains a specialized synaptic extracellular matrix that influences the organization of pre and post synaptic elements, and regulates synapse function (for example ach esterase and agrin at the nmj). Acetylcholine (ach) is the transmitter at the mammalian nmj. In our invertebrate it is glutamate. Maturation of the postsynaptic apparatus method label achrs with floro bungarotoxin. Before motoneuron innervation achrs form loose aggregates and are distributed across the surface of the myotube. Following innervation, the receptors cluster and the distribution becomes increasingly complex as the synapse matures. The distribution of receptors matches the presynaptic input.
