Document Summary

Gaba receptors and their synapses ii: dynamics of inhibition in health and disease. Inhibition in health: phosphorylation of gabar, scaffolding protein: gephyrin. Important for inhibitory neurotransmitter receptors: receptor trafficking. Inhibition in disease: gephyrin as a signaling hub in homeostatic plasticity, dysregulation of nkcc1 and kcc2 in pain, neurosteroids in stress, gabaar in idiopathic generalized epilepsy. The life cycle of the gaba-a receptor complex is regulated by protein kinases. Pka, pkc and src kinases are involved in regulation of gabar. Camkii is important regulator of gabaergic synapses. Gabar phosphorylation is tied into endocytosis or presentation on the synapse. Gaba-a receptor signaling strengthened by phosphorylation of beta and gamma subunits. Pption changes synaptic event amplitude and/or decay kinetics. Camkii is pivotal in regulating glur, activated by elevated ca2+ intracellularly mediated by nmdar, phosphorylate ampar, promote insertion of ampar into glutaminergic synapses. Increased ca2+ inside cell activates camkii which phosphorylates beta or gamma subunit of synaptic gabar and this causes 2 functional consequences.
