It is necessary for specialized structures like microtubules in cilia and actin in microvilli. The cytoskeleton must be dynamic for cell movement (visualized with actin-gfp) and cell division (visualized with tubulin-gfp). Cilia = microtubules and microvilli = actin. Defined based on their diameter and monomers used to build the filament: actin can be tagged with fluorescently- tagged phalloidin molecule (derived from death cap mushroom). Phalloidin: has high affinity and specificity to the actin monomers and it also stabilizes the filament. Actin can also be labeled with an actin antibody or with actin-gfp: microtubules labeled with antibodies. Intermediate filaments (ifs) labeled with antibodies specific to monomers, or gfp-fusion. Made of actin-subunits which join to make actin polymer. There is different distribution of the filaments in different cells. Example in epithelial cell the actin fibers form microvilli at cell surface microtubules are used for intracellular transport intermediate filaments help with structure.
