BIOLOGY 2B03 Lecture Notes - Lecture 2: Hydrolysis, Peptide, Atp Hydrolysis
Document Summary
When else do proteins unfold/misfold: heat shock protein expression increases during aging (fruit flies, mice) and in mouse model systems of degenerative disorders (huntington"s and alzheimer"s diseases) Hsps protect against stress: when more favourable conditions return, the hsps are again down-regulated. Hsps increase survival: take away the hsp70 and survival rate is lower, add an extra copy of hsp70 and survival increases. Protein folding: what we believe happens: in theory, there are rules for folding a protein into the lowest energy stable state, what really happens: in the cell, internal and external stressors that promote unfolding and misfolding. Thermodynamic hypothesis: or anfinsen"s dogma, due to thermodynamic molecular forces, polypeptides automatically assume unique, stable conformations, every sequence of amino acids auto-assembles into a unique, defining conformation, exceptions exist. 1 and 2 are not the same things. *basically saying that proteins will spontaneously go into their native state.
