Document Summary

1: select protein source, extract the proteins break open cell, solubilize protein, stabilize protein, fractionate, determine purity. Method of detecting for the presence of a specific protein and estimating the concentration of the protein. Should be specific to the protein you are studying and based upon unique characteristics: ex. Source: protein should be easily obtained and present in large amounts: cell low in proteins that may co-purify, low in proteases that could destroy your protein or add protease inhibitors. Predict whether you expect a protein to be soluble based upon the properties that you believe the protein has: ex. Soluble proteins = cytosolic/secreted proteins: insoluble proteins = transmembrane/membrane-associated proteins. Can increase solubility of protein by changing some conditions in your extract. Affected by solution ph, salt concentration: detergents can increase solubility of insoluble proteins in solution. Fractionation: process of separating proteins into different groups or fractions. Proteins vary in size, polarity, charge, solubility and shape.
